Patent number: 8895292Abstract: A microfluidic chip device (MCD) and its use for performing miniaturized assays on magnetic microbeads (MMs) are described. The MCD is particularly useful for carrying out miniaturized transcript analysis by aiding affinity capturing (TRAC) assays, including PCR. The MCD comprises at least one reaction chamber with sealable liquid connections and at least one fluidic pillar filter in each chamber. The fluidic pillar filter comprises rods with spacings allowing MMs to pass. The sealable liquid connections feed liquid to the reaction chamber, wherein air bubbles are removed. The liquid stream contacts the MMs, which are manipulated with a magnetic rod. The liquid connections enable trapping of the MMs behind the pillar filters or in the channel, while the liquid is changed.Type: GrantFiled: April 9, 2009Date of Patent: November 25, 2014Assignee: Valtion Teknillinen TutkimuskeskusInventors: Hans Söderlund, Ari Hokkanen, Kari Kataja, Ingmar Stuns, Kai Kolari, Heli Siren, Stella Rovio, Reetta Satokari, Jan Rautio

Patent number: 8445426Abstract: The present invention relates to the fields of life sciences and food, feed or pharmaceutical industry. Specifically, the invention relates to novel peptides, pilus structures, polynucleotides as well as vectors, host cells, products and pharmaceutical compositions comprising the polynucleotides, peptides or pilus structures. The invention also relates to gene clusters and antibodies. Furthermore, the present invention relates to methods for producing the peptides or pilus structures or producing the products comprising the peptides or pilus structures. Furthermore, the present invention relates to treatments as well as uses and methods for screening bacterial strains, for reducing or inhibiting the adhesion of pathogenic bacteria, promoting the adhesion of bacterial cells to the mucus and for modifying immune response in a subject. Still, the present invention relates to methods for detecting probiotic bacterial strains or pathogen strains.Type: GrantFiled: February 2, 2009Date of Patent: May 21, 2013Assignee: Valio Ltd.Inventors: Willem Meindert De Vos, Airi Palva, Ilkka Palva, Justus Reunanen, Ingemar Von Ossowski, Reetta Satokari, Satu Vesterlund, Matti Kankainen, Tuomas Salusjärvi, Soile Tynkkynen

Patent number: 8143388Abstract: The invention relates to a method and test kit for quantitative determination of the amounts or relative proportions of polynucleotides in a mixture. The invention enables assessment of dynamic variations in a mixed population of organisms using affinity aided solution hybridization. The test kit comprises organized pools of polynucleotide probes having approximately the same number of nucleotides, which are distinguishable using resolution enabling tags providing the probes with different sizes. The resolution enabling tags may simultaneously act as tracer, affinity or primer tags. The probes are allowed to hybridize with affinity tagged analyte polynucleotides. The result is hybrids, recoverable on separation aiding tools provided with counterparts of the affinity tag. After the quantitative release of the probes, the individual probes can be amplified and recorded.Type: GrantFiled: April 18, 2008Date of Patent: March 27, 2012Assignee: Valtion Teknillinen TutkimuskeskusInventors: Hans Söderlund, Reetta Satokari, Kari Kataja, Kristiina Takkinen
