  • Patent number: 5480805
Abstract: A structural gene encoding a polypeptide with .DELTA.8-7 sterol isomerase activity is disclosed, Recombinant DNA molecules useful for transforming yeast and mutant yeast transformed with such recombinant DNA molecules are also disclosed. The structural gene is useful for modulating the accumulation of sterols in yeast comprising increasing the expression level of a structural gene encoding a polypeptide having .DELTA.8-7 sterol isomerase activity in a mutant yeast having single or double defects in the expression of sterol biosynthetic enzymes is provided.
    Type: Grant
    Filed: May 10, 1994
Date of Patent: January 2, 1996
    Assignee: Amoco Corporation
Inventors: Fred R. Wolf, Richard E. Cuellar
  • Patent number: 5365017
Abstract: A method of increasing sterol accumulation in a plant by increasing the copy number of a gene encoding a polypeptide having HMG-CoA reductase activity is disclosed. The copy number is preferably increased by transforming plants with a recombinant DNA molecule comprising a vector operatively linked to an exogenous DNA segment that encodes a polypeptide having HMG-CoA reductase activity, and a promoter suitable for driving the expression of said polypeptide. Also disclosed are a method of increasing cycloartenol accumulation in a plant, a method of increasing the resistance of plants to pests and the transformed plants themselves.
    Type: Grant
    Filed: September 9, 1993
Date of Patent: November 15, 1994
    Assignee: Amoco Corporation
Inventors: Joseph Chappell, Court A. Saunders, Fred R. Wolf, Richard E. Cuellar

  • Patent number: 5137817
Abstract: An electrode set for electrotransformation of a host cell by electroporation, comprising: an electrically insulating substrate; and at least one pair of interdigitated electrodes which are carried by said substrate and which lie in a generally planar array, said electrodes and said substrate being adopted to carry a cell transformant and a host cell thereon, said electrodes and said host cell and said cell transformant being adapted to pass electrical current upon the application of a potential difference across said electrodes.
    Type: Grant
    Filed: October 5, 1990
Date of Patent: August 11, 1992
    Assignee: Amoco Corporation
Inventors: Heinz H. Busta, Michael L. Bittner, Richard E. Cuellar
  • Patent number: 4886878
Abstract: Modified 19kd and 22kd zeins containing lysine are provided. Lysine additions are made by modifying structural genes which encode 19kd and 22kd pre-zeins. Modified zeins produced from the modified structural gene templates are found to retain important properties of unmodified zeins, having similar molecular weights, having the ability to form protein bodies within the rough endoplasmic reticulum of a host cell and being soluble in alcohol.
    Type: Grant
    Filed: November 22, 1988
Date of Patent: December 12, 1989
Assignees: Lubrizol Genetics, Inc., Purdue Research Foundation, Inc.
Inventors: Brian Larkins, Richard E. Cuellar, John C. Wallace, Gad Galili
