    Abstract: A method for determining the sex of a mammalian embryo is disclosed. This method begins with the isolation of an embryonic cell that contains pseudo-autosomal DNA sequences. At least a portion of the pseudo-autosomal DNA sequences are amplified, creating pseudo-autosomal DNA fragments. Allele-specific DNA sequences are amplified using the pseudo-autosomal DNA fragments as a template and allele-specific primers. The allele-specific products are examined for the presence or absence of the X-allele-specific and Y-allele-specific products. Preferably, the embryo is bovine and the pseudo-autosomal DNA sequences are ZFX and ZFY. Also preferably the allele-specific primers are those described in Sequence ID NOS: 5, 6, 7 and 8.
