  • Publication number: 20160123856
Abstract: Apparatus and methods are provided to obtain RNA-enhanced and protein-enhanced fluid samples which are stable at ambient temperatures. An apparatus includes a filter element made from a fibrous hydrophilic material which preferentially binds and filters cells, DNA-containing macrostructures, mucins and particulates, but does not preferentially bind RNA, RNA-containing macrostructures or proteins. An apparatus may include a sample collector having an absorbent collection pad which also does not preferentially bind RNA, RNA-containing macrostructures or proteins. A method includes obtaining a sample and passing the sample through an RNA-and-protein-preferential filter element to obtain RNA-enhanced and protein-enhanced samples. The method may include use of a RNA-and-protein-preferential absorbent pad material to collect the sample. DNA-rich samples may be obtained by using DNA elution buffer to release the DNA-containing cells and macrostructures after obtaining the RNA-enhanced and protein-enhanced samples.

  • Publication number: 20090253219
Abstract: Methods and devices for the detection and/or quantification of an analyte in a sample are provided. These are positive detection methods and devices, in that the more analyte is present in the sample, the stronger the signal that is provided. Devices of the invention include a mobilization zone including a mobile or mobilizable detectable analyte analog, a sample application area, primary and secondary capture areas each including an immobilized binding partner having a binding affinity for the analyte being tested for a detectable analyte analog. The mobilization zone, sample application area, primary and secondary capture area are in fluid continuous contact with each other. In these devices, the first immobilized binding partner has an equal or lower apparent affinity for the analyte than it has for the detectable analyte analog.

  • Publication number: 20090246886
Abstract: A lateral flow test strip is disclosed that includes a path of flow from a sample receiving zone through a mobilization zone to a primary capture zone and a secondary capture zone. A mobilizable conjugate is present in the mobilization zone, and a mobilizable label is present on the test strip upstream of the conjugate. An immobilized first specific binding partner is present in the primary capture zone and an immobilized second specific binding partner is present in the secondary capture zone. The conjugate includes a primary specific binding partner for the first specific binding partner in the primary capture zone, and a secondary specific binding partner that binds the label and the second specific binding partner. Application of liquid sample to the sample receiving zone results in movement of the liquid sample along the path of flow to move the label and conjugate distally along the test device.
