    Abstract: The present invention provides engineered fatty acid desaturase molecules preferring Gamma Linolenic Acid (GLA) over Linoleic Acid (LA) as a substrate. The invention further discloses compositions, polynucleotide constructs, transformed host cells, transgenic plants and seeds comprising the desaturase molecule, and methods for preparing and using the same. In particular, the disclosed engineered desaturase molecules are capable of altering the omega-3 fatty acid profiles in plants and plant parts.

Sequences of the loci are useful for designing primers and probe oligonucleotides for detecting polymorphisms in soybean DNA. Polymorphisms are useful for genotyping applications in soybean. The polymorphic markers are useful to establish marker/trait associations, e.g. in linkage disequilibrium mapping and association studies, positional cloning and transgenic applications, marker-aided breeding and marker-assisted selection, and identity by descent studies. The polymorphic markers are also useful in mapping libraries of DNA clones, e.g. for soybean QTLs and genes linked to polymorphisms.

    Abstract: Systems are disclosed that are useful for introducing one or more targeted positions or regions of diversity into a nucleic acid molecule. In certain embodiments, diversity in a targeted position or region is generated by providing one or more degenerate primer sets and a template nucleic acid molecule, wherein the primers are extended in opposite directions against the template nucleic acid molecule in a polymerase-mediated extension reaction. In certain embodiments, the generated nucleic acid molecule into which diversity has been introduced comprises single-stranded regions at its termini, which single-stranded regions are capable of annealing to each other.
