  • Publication number: 20250122482
Abstract: The present inventions provide covalently surface modified adeno-associated viruses can comprise gene of interest (GOIs) and advantageously can be targeted to certain cell and tissue types for preventative and therapeutic purposes. The present inventions further provide systems and methods for engineering adeno-associated virus (AAV) to create covalently surface modified adeno-associated viruses, and methods of purifying such covalently surface modified adeno-associated viruses. The inventions further include covalently surface modified adeno-associated viruses and preparations and products comprising such covalently surface modified adeno-associated viruses.

  • Publication number: 20240425825
Abstract: The inventions provide methods of purifying full adeno-associated virus (AAV) capsids, wherein the method comprises the steps of (a) subjecting a sample comprising AAV capsids in buffer to anion exchange chromatography (AEX) to separate full AAV capsids from capsids that are not full (that is, partially-filled AAV capsids, nearly empty AAV capsids and empty AAV capsids) and form a first pool (“first AEX pass”), wherein the first pool is enriched in the ratio of full capsids to capsids that are not full; and (b) subjecting the first pool to anion exchange chromatography to form a second pool (“second AEX pass”), wherein the second pool is further enriched in the ratio of full capsids to capsids that are not full (that is, partially-filled AAV capsids, nearly empty AAV capsids and empty AAV capsids), wherein the enrichment in full capsids can be least 3 fold. Third, fourth and more AEX passes also can be undertaken.

  • Publication number: 20050267271
Abstract: Bimetallic catalyst for producing polyethylene resins with a bimodal molecular weight distribution, its preparation and use. The catalyst is obtainable by a process which includes contacting a support material with an organomagnesium component and carbonyl-containing component. The support material so treated is contacted with a non-metallocene transition metal component to obtain a catalyst intermediate, the latter being contacted with an aluminoxane component and a metallocene component, This catalyst may be further activated with, e.g., alkylaluminum cocatalyst, and contacted, under polymerization conditions, with ethylene and optionally one or more comonomers, to produce ethylene homo- or copolymers with a bimodal molecular weight distribution and improved resin swell properties in a single reactor. These ethylene polymers are particularly suitable for blow molding applications.

  • Publication number: 20050054791
Abstract: An ethylene/?-olefin copolymer comprising a component produced by a non-single-site polymerization catalyst and a component produced by a single-site polymerization catalyst, its preparation and use are described. The copolymer has an ?-olefin content of 5 to 20 percent by weight aud shows at least two CRYSTAF peak temperatures differing by at least 15° C. and/or at least two DSC melting peak temperatures differing by at least 15° C.

  • Publication number: 20050003950
Abstract: Methods of making mixed Ziegler-Natta/metallocene catalysts are disclosed. The methods include slurrying a dehydrated support in a hydrocarbon solvent, adding a dialkylmagnesium compound, then adding a non-metallocene group 4 or 5 metal compound, followed by adding a metallocene, an alumoxane, and optionally an aklyl aluminum compound and an electron donor compound, followed by drying the product. The alumoxane is added in an aromatic solvent.

  • Patent number: 4312782
Abstract: A novel catalyst system for the polymerization of alpha-olefins is provided. The catalyst system comprises: (a) an organoaluminum containing component, e.g. triethyl aluminum, and (b) a titanium halide containing component. The titanium halide containing component is obtained by copulverizing a halogen containing magnesium compound, e.g. MgCl.sub.2, with an active hydrogen containing organic compound, e.g. phenol, to produce a copulverized product. The copulverized product is then reacted with an organometallic compound of a metal of Groups I to III of the Periodic Table, e.g. triethyl aluminum, to produce a reaction product. The reaction product is then copulverized with a complex of a first titanium halide compound and an electron-donor, e.g. TiCl.sub.4.ethyl benzoate, to produce a solid reaction product. The solid reaction product is then reacted with a second titanium halide compound, e.g. TiCl.sub.4.
