Abstract: Efforts to use RNA-cleaving DIMA enzymes (DNAzymes) as gene silencing agents in therapeutic applications have stalled due to their low efficacy in clinical trials. Here the present invention reports a xeno-nucleic acid (XNA) modified version of the classic DNAzyme 10-23 that achieves multiple turnover activity under cellular conditions and resists nuclease digestion. The new reagent overcomes the problem of product inhibition limiting previous 10-23 designs using molecular chemotypes with DNA. FANA, and TNA backbone architectures that balance the effects of enhanced biological stability with RNA hybridization and divalent metal ion coordination. In cultured mammalian cells. X 10-23 facilitates persistent gene silencing by efficiently degrading exogenous and endogenous mRNA transcripts. Together, these results demonstrate that new molecular chemotypes can improve the activity and stability of DNAzymes, and may provide a new route for nucleic acid enzymes to reach the clinic.

  • Publication number: 20240390977
Abstract: A device for production of heavy metal powders by ultrasonic atomization from a heavy metal raw material is provided. The device comprises a feeding means, a heat source, an adjusting means, and a collecting means. The feeding provides the heavy metal raw material in the vicinity of the heat source, and the heat source generates an electric arc to heat the heavy metal raw material to create a molten metal pool on a sonotrode. The sonotrode provides ultrasonic mechanical vibrations to the molten metal pool to cause heavy metal droplets to be ejected from the molten metal pool. The adjusting means adjusts the feeding means, the heating means, and the sonotrode to direct the heavy metal droplets to cause the heavy metal droplets to freely cool down within a predetermined distance and transform the heavy metal droplets to the heavy metal powder.

  • Publication number: 20240397359
Abstract: A method of assessing latency in a communications network comprising attempting to contact from a UE in a cellular network a specific internet resource with a HTTP request addressed to the resource and capturing data specifying the period between making the attempt and receiving at the UE from the resource a response to the request. Thus, data can be obtained from which can be made an end-to-end measurement of the latency affecting a user of a UE. Such measurements can be used by the operator of the communications network to, amongst other things, assess whether the network is performing satisfactorily, to advise users about the performance of the network and, determine that hardware or software within the network needs repair, upgrade, or augmentation.
