    Abstract: A method of generating corrected fluorescence data of concentrations of a targeted fluorophore in tissue of a subject includes administering first and second fluorescent contrast agents to the subject, the first contrast agent targeted to tissue of interest, the second agent untargeted. The tissue is illuminated with light of a first stimulus wavelength and first data is acquired at an appropriate emissions wavelength; the tissue is illuminated at a second stimulus wavelength and second data is acquired at a second emissions wavelength associated with the second agent, the first and second emissions wavelength differ. Difference data is generated by subtracting the second data from the first data. A system provides for stimulus and capture at multiple wavelengths, with image storage memory and subtraction code, to perform the method. Corrected data may form an fluorescence image, or is used to generate fluorescence tomographic images.

    Abstract: A processor receives a first image of a first section of a specimen contacted with at least one first fluorophore having an emission spectrum in the range of approximately 200 nm to 1000 nm, the first image created using a first wavelength of invisible light. The processor receives a second image of a second section of the specimen, the second section adjacent to the first section and the second image created using the first wavelength of invisible light. The processor co-registers the first image and the second image. The processor creates a single-plane image of the first section using a next-image process; wherein the creating of the image includes a debluring process. The processor receives an image of a third section of the specimen created using the first wavelength of invisible light. The processor creates a 3D image of the specimen using at least three images of adjacent specimen sections.

    Abstract: An improved pipette probe in an automated chemical analysis system is provided, along with a pipette tip wash protocol to reduce carryover. The pipette tip comprises an elongated stainless-steel barrel which houses a polytetrafluoroethylene tube. The non-wettable properties of polytetrafluoroethylene inhibit fluid adhesion and thereby reduce carryover. The pipette tip is swaged at a fluid dispensing end to a restrictive diameter for good pipetting precision and accuracy. A novel wash protocol is also provided which includes a novel wash solution comprising quantities of Triton X-100 nonionic detergent, Diethanolamine, and NaCl.
