  • Patent number: 11442064
Abstract: The present invention includes a novel method and system for identification of microorganisms in samples that proteins and other biological material from non-microorganism sources (e.g., proteins of mammalian origin) that can interfere with identification of the microorganisms. The methods and systems described herein include use of a single-use chromatography medium to purify intact proteins prior to mass spectrometry analysis. The chromatography medium and the methods described herein can rapidly and efficiently remove of a substantial portion of interfering biological material (e.g., mammalian proteins) from a crude cell lysate while preserving high signal strength and removing enough of the interfering protein(s) to allow for identification of the microorganism(s) by mass spectrometry analysis.
    Type: Grant
    Filed: August 24, 2017
Date of Patent: September 13, 2022
    Assignee: Thermo Fisher Scientific OY
Inventors: Mikko Koskinen, Leena Valmu, Joni Rannisto, Kelly Flook, Roger Grist, Anssi Rantakari

  • Publication number: 20190006161
Abstract: Methods and systems for identification of microorganisms either after isolation from a culture or directly from a sample. The methods and systems are configured to identify microorganisms based on the characterization of proteins of the microorganisms via high-resolution/mass accuracy single-stage (MS) or multi-stage (MSn) mass spectrometry. Included herein are also discussion of targeted detection and evaluation of virulence factors, antibiotic resistance markers, antibiotic susceptibility markers, typing, or other characteristics using a method applicable to substantially all microorganisms and high-resolution/mass accuracy single-stage (MS) or multi-stage (MSn) mass spectrometry.
