    Abstract: An embodiment provides a method for measuring an analyte of a sample using a colorimeter, including: introducing the sample into a measurement chamber of the colorimeter, wherein the measurement chamber comprises an optical path from a light source to an optical sensor; identifying and waiting a sample settling period, wherein the sample settling period comprises a dynamic length of time, wherein the identifying comprises: measuring, using the optical sensor, a light throughput of the sample; and determining, based upon the measuring, the light throughput has reached a stable baseline; and obtaining a reference measurement of the sample when the stable baseline is reached. The server may determine that the first request is associated with a higher priority than the second request and, in response, may reserve the first number of cores for the first amount of time for the first application.     Abstract: Compartment-free single cell genetic analysis methods are provided. Aspects of the methods include: (a) combining a cellular sample with a plurality of distinct barcoded beads comprising barcoded reverse primers under conditions sufficient to produce a liquid composition comprising a plurality of separated cell/barcoded bead complexes; (b) hybridizing template binding domains of barcoded reverse primers to template nucleic acids of the cells to produce primed template nucleic acids; and (c) subjecting the primed template nucleic acids to primer extension reaction conditions sufficient to produce barcoded nucleic acids, e.g., for subsequent amplification and analysis, such as by Next Generation Sequencing (NGS) protocols.
