  • Publication number: 20070054322
Abstract: The present invention relates to a method for the diagnosis of a neurodegenerative disorder in a mammalian subject. The method of the invention comprises the steps of (a) providing a body fluid sample of said subject; (b) concentrating proteins comprised within said sample by a suitable means; (c) contacting the concentrated sample obtained in step (b) with a sufficient amount of a protein which has a beta-sheet structure, preferably, an IgG light chain, under conditions suitable to allow the formation of aggregates. Which aggregates comprise a protein associated with a neurodegenerative disorder, and (d) measuring aggregate formation by suitable means. Whereby the presence of aggregates in said sample indicates that said subject carries said neurodegenerative disorder. The invention further provides for kits and diagnostic compositions for the detection of a neurodegenerative disorder, particularly, prion diseases.

  • Publication number: 20050222275
Abstract: The present invention relates to a pharmaceutical composition for the treatment and/or delaying the onset of clinical symptoms and/or delaying the progress of a neurodegenerative disorder caused by prions. The composition of the invention comprises as an active ingredient DMSO, and optionally further comprising pharmaceutically acceptable additives and/or diluents. The composition of the invention is particularely applicable in the treatment of neurodegenerative disorders associated with enhanced accumulation of PrPSc into amyloid plaques. The invention further relates to methods of treatment of a neurodegenerative disorder caused by prions and to the use of DMSO in the preparation of a pharmaceutical composition for the treatment and/or delay of the onset of clinical symptoms and/or delay of progress of neurodegenerative disorders caused by prions.

  • Publication number: 20050084983
Abstract: The present invention relates to a method for detecting the presence of the abnormal isoform of prion protein (PrPSC) in a urine sample of a subject. The method of the invention comprising the steps of: (a) providing a urine sample of said subject; (b) isolating from said sample all proteins, preferably, isolating proteins having a molecular weight higher than about 8 Kda; (c) optionally, and preferably, subjecting the proteins obtained in step (b) to protease digestion, and isolating from the mixture obtained in step (c) any protease resistant proteins; and (d) detecting the presence of PrPSC in the protease resistant fraction obtained in step (c) by a suitable detection technique. Furthermore, the invention further relates to methods for diagnosing a prion disease in a subject and for screening donors of blood samples for the presence of prion diseases. The invention further provides for a diagnostic kit for diagnosing a prion disease in a subject.
