    Abstract: Provided herein are compositions and systems comprising engineered cells useful for the measurement of autophagy induction, maturation and flux in a high-throughput manner. In particular, provided herein are human embryonic kidney (HEK) cells that express the native autophagy substrate and marker protein LC3 fused to Dendra2, a photoconvertable fluorescent protein; this manipulation allows assessment of all three stages of autophagy (e.g., induction, maturation, and cargo degradation), noninvasively in living cells, without the need for protein overexpression that may interfere with autophagy activity.
