Publication number: 20240127451Abstract: Described herein are methods and computer systems for classification of CD8 T-cell topology using a patient response-based linear cutoff model. A plurality of histology images of tissue samples in a plurality of patients are received by a computer system. An image analysis of the plurality of histology images is performed to obtain a CD8+ T-cell abundance in the tumor parenchyma and stroma in each of the plurality of histology images. Real inflammation scores and tumor infiltration scores are determined based on a polar coordinate transformation of the CD8+ T-cell abundance in the tumor parenchyma and stroma. Based on the real inflammation scores and tumor infiltration scores, a feature space is generated, and linear boundaries or linear cutoffs between a plurality of classifications in the feature space are identified based on the real inflammation scores, the tumor infiltration scores, and patient response data.Type: ApplicationFiled: February 25, 2022Publication date: April 18, 2024Applicant: Bristol-Myers Squibb CompanyInventors: George C. Publication number: 20230326022Abstract: A method includes receiving an input histology image, processing, using a cell classification model, the input histology image to generate one or more lymphocyte density maps within the input histology image, and performing morphological image processing on the one or more lymphocyte density maps to identify one or more TLS regions within the input histology image. Each TLS region is represented by a respective cluster of lymphocyte cells. For each corresponding TLS region of the one or more TLS regions identified in the input histology image, the method also includes extracting, from the respective cluster of lymphocyte cells, a respective set of TLS features, and processing, using a TLS classification model, the respective set of TLS features to classify the corresponding TLS region as one of a first TLS maturation state, a second TLS maturation state, or a third TLS maturation state.Type: ApplicationFiled: April 7, 2023Publication date: October 12, 2023Applicants: Bristol-Myers Squibb Company, PathAI, Inc.Inventors: Vanessa Matos-Cruz, George Lee, Varsha Chinnaobireddy, Maryam Pouryahya, Darren Thomas Fahy, Christian Winskell Kirkup, Kathleen Sucipto, Sai Chowdary Gullapally, Archit Khosla, Nishant Agrawal, Benjamin Patrick Glass, Sergine Brutus, Limin Yu, Murray Berle Resnick, Rachel L. Publication number: 20230306762Abstract: Described herein are methods and computer systems for classification of CD8 T-cell topology using artificial intelligence and machine learning. A plurality of histology images of tissue samples in a plurality of patients are received by a computer system. An image analysis of the plurality of histology images is performed to obtain a CD8+ T-cell abundance in the tumor parenchyma and stroma in each of the plurality of histology images. A machine learning algorithm is then trained using results of the image analysis and the CD8+ T-cell abundance in the tumor parenchyma and stroma. Based on the training, a machine learning feature space comprising a plurality of classifications is generated, and boundaries between the plurality of classifications in the machine learning feature space are identified.Type: ApplicationFiled: August 31, 2021Publication date: September 28, 2023Applicant: Bristol-Myers Squibb CompanyInventors: George C. Patent number: 8608780Abstract: A spinal implant provides support for desired parts of the spine. The spinal implant includes a pair of elongated members and a variable length cross-link. A variable length cross-link device may include a fixed portion having a receiver portion for attachment to a first elongated member, a transverse portion ,and an adjustable portion having a receiver portion for attachment to a second elongated member and a transverse portion engaging member. Inserting the transverse portion of the fixed portion into the engaging portion of the adjustable portion may form a cross-link for stabilizing motion between two elongated members. Engaging the adjustable portion at a selected point on the transverse portion establishes a length selected by the surgeon. The surgical procedure may use minimally invasive surgery or non-minimally invasive surgery, as desired. Components may be inserted through sleeves attached to various coupling devices, or may be inserted and guided along wires.Type: GrantFiled: October 4, 2011Date of Patent: December 17, 2013Assignee: Zimmer Spine, Inc.Inventors: Charles R. Patent number: RE46379Abstract: The present invention relates to a method of determining the presence of a retinoblastoma protein (Rb)-mediated cancerous or pre-cancerous condition in a sample of fixed cells that involves exposing the sample to an antibody that recognizes phosphorylation of serine on the retinoblastoma protein (pS-Rb), and identifying cells in the sample that are recognized by the antibody to pS-Rb, where the recognition of pS-Rb in the sample determines the presence of an Rb-mediated cancerous or pre-cancerous condition. The method also involves identifying the presence of specific cell cycle markers and positive and negative cell cycle regulators to assess the stage of disease progression in a cancer patient having an (Rb)-mediated cancer. The present invention also relates to methods of assessing the efficacy of cancer therapy for an Rb-mediated cancer, determining the mechanism of cell cycle progression in an Rb-mediated cancer, and identifying cells predisposed to developing an Rb-mediated cancerous condition.Type: GrantFiled: December 9, 2015Date of Patent: April 25, 2017Assignee: Cornell Research Foundation, Inc.Inventors: Selina Chen-Kiang, Scott A.
