  • Patent number: 11821000
Abstract: A method of separating a liquid sample containing viral vectors includes flowing the liquid sample into an anion exchange column. A first viral vector contains a targeted genetic material and a second viral vector contains essentially no genetic material or a non-targeted genetic material, wherein the targeted genetic material is different than the non-targeted genetic material. A mobile phase can be flowed into the anion exchange column, wherein the mobile phase includes a buffer solution A and a buffer solution B. The buffer solutions A and B both include volatile buffer salts. The first viral vector and the second viral vector can be separated so that the first viral vector and the second viral vector elute off the anion exchange column at different times and then be detected.

  • Patent number: 11022585
Abstract: A plurality of chromatograms is run using a salt gradient with a range of constant pH values or using a pH gradient with a range of constant salt concentrations. A chromatography optimization algorithm can be used to identify at least one salt gradient chromatogram or at least one pH gradient chromatogram to establish optimized buffer conditions. The chromatography optimization algorithm can include a total number of peaks algorithm, a peak-to-valley algorithm, and/or a peak capacity algorithm.

  • Publication number: 20190358621
Abstract: A chromatographic media for separating bio-polymers, the chromatographic media having cationic exchange properties and anionic exchange properties, the chromatographic media comprising: (a) non-porous substrate particles including an organic polymer, the substrate particles having a neutral hydrophilic layer at a surface of the non-porous substrate particles, in which the neutral hydrophilic layer is configured to reduce a binding of the bio-polymers directly to the non-porous substrate particles compared to a binding of the bio-polymer to the non-porous substrate particles without the neutral hydrophilic layer; (b) a charged first ion exchange layer bound to the substrate particles on top of the hydrophilic layer, the first ion exchange layer comprising first ion exchange groups; and (c) a charged second ion exchange layer bound to the substrate particles on top of the first ion exchange layer.

  • Patent number: 8921113
Abstract: A buffer kit includes a first eluent and second eluent. The first eluent solution includes at least four buffer salts where at least three of the four buffer salts are a monovalent buffer salt, have a net negative charge or a net neutral zwitterionic charge, and include a sulfonate group and an amine. The second eluent solution includes at least four buffer salts where at least three of the four buffer salts are a monovalent buffer salt, have a net negative charge or a net neutral zwitterionic charge, and include a sulfonate group and an amine. The first eluent solution has a first pH and the second eluent solution has a second pH where the first pH and second pH are different values. The buffer kit provides a linear pH gradient that forms an approximately straight line from at least the first pH to the second pH.
