  • Publication number: 20140154783
Abstract: The present invention relates to molecular biology, molecular genetics, and bioprocessing. The embodiments provide for compositions and methods for producing a biological product, such as an immunogenic agent, in an embryonated egg by introducing into the egg a RNA effector molecule capable of modulating expression of a target gene, wherein the modulation enhances production of the biological product in the egg. These methods provide for RNAi-based approaches to optimize the production of biologics from embryonated eggs, such as the production of viral vaccines including seasonal and pandemic flu vaccines. The invention also relates to molecules, reagents, cells, and kits useful for carrying out the methods, and biological products produced by the methods.

  • Publication number: 20140099666
Abstract: The invention provides compositions and methods for producing a biological product from a host cell. In various embodiments, the biological product is a polypeptide, a metabolite, a nutraceutical, a chemical intermediate, a biofuel, a food additive, or an antibiotic. In one aspect, the invention provides for a method for producing a biological product from a host cell. The method generally comprises contacting the cell with a RNA effector molecule, a portion of which is complementary to a target gene, maintaining the cell in a large-scale bioreactor for a time sufficient to modulate expression of the target gene, wherein the modulation enhances production of the biological product from the cell, and isolating the biological product from the cell.

  • Patent number: 8557743
Abstract: Disclosed are methods of selecting phage encoding a target binding protein. The methods can include forming a mixture comprising a plurality of diverse display phage, a target, and a support, and forming phage immobilized to the support, each of which comprises a phage which binds the target and the target immobilized to the support. Phage that do not bind to the target are separated. Host cells are contacted with the phage immobilized to the support via binding to the target so that the host cells are infected by the phage immobilized to the support. Replicate phage are produced from the infected cells in the presence of the target immobilized to the support, thereby forming replicate phage immobilized to the support via binding to the support. Replicate phage that do not bind to the target are separated. Host cells are contacted with the replicate phage immobilized to the support.

  • Publication number: 20050009136
Abstract: The invention provides, inter alia, proteins that bind to PAPP-A, an 1547 amino acid glycoprotein which can form an ˜200 kDa monomer or an ˜400 kDa dimer. In one form, the proteins are antibodies. In one embodiment, the proteins can inhibit the ability of PAPP-A to interact (e.g., cleave) substrates such as IGFBP-4, IGFBP-5, and IGFBP-2.

  • Publication number: 20030170686
Abstract: The invention features apparati and methods for washing magnetically responsive particles such as paramagnetic beads. One exemplary apparatus includes a flow chamber having an inlet and an outlet; and at least a first magnetic field inducer. The apparatus is configured such that a first magnetic field can be selectively applied in the flow chamber. The apparatus can also apply a flow of fluid to the chamber in coordination with the selective application of the magnetic field.
