  • Patent number: 10443026
Abstract: A polygonal scaffold includes at least three polyproline II (PPII) helix rods and at least three connectors. Each of the PPII helix rods is composed of a plurality of repeat units represented by Formula (i), and at least one of the PPII helix rods has at least one hydrogen atom of at least one of the repeat units being substituted by a first chemical handle for connecting a ligand. Each of the connectors is a divalent organic group, and every two of the PPII helix rods are connected by one of the connectors to form a closed ring. Therefore, with the uniform composition, the PPII helix rods have a stable helical structure under aqueous conditions, and a desired rigidity can be provided.

  • Publication number: 20190079086
Abstract: A detection structure including a substrate, a coating structure and a plurality of polyproline helix structure is provided. The substrate has a plurality of detecting regions. The coating structure is located on the substrate. The plurality of polyproline helix structure is located in each of the detecting region and on the coating structure. Each of the polyproline helix structures is composed of a plurality of proline monomers aligned in the first direction, aligned in the second direction and aligned in the third direction. The proline monomer aligned in the first direction is connected in the coating structure through a connecting structure, and the proline monomer aligned in the second direction is connected to at least two ligands. The two ligands on each of the polyproline helix structure has a fixed distance that can be adjusted. A production method of the detection structure above is also described.

  • Publication number: 20090232831
Abstract: A conserved cluster of oligomannose glycans on gp120 has been identified as the epitope recognized by the broadly HIV-1-neutralizing monoclonal antibody 2G12. Oligomannose glycans are also the ligands for DC-SIGN, a C-type lectin found on the surface of dendritic cells. Multivalency is fundamental for carbohydrate-protein interactions, and mimicking of the high glycan density on the virus surface has become essential for designing carbohydrate-based HIV vaccines and antiviral agents. Synthesis of oligomannose dendrons, which display multivalent oligomannoses in high density, and characterize their interaction with 2G12 and DC-SIGN by a glycan microarray binding assay is disclosed. These glycodendrons inhibit the binding of gp120 to 2G12 and recombinant dimeric DC-SIGN with IC50 in the nanomolar range. A second-generation Man9 dendron was identified as a potential immunogen for HIV vaccine development and as a potential antiviral agent.
