Once the range of dimensions is determined, a light absorbent composition can be found which absorbs light at a range of wavelengths that at least substantially covers the range of dimensions of the domains. This material can then be soaked and soaked liquid filtered by nano-filtration. When nano-filtration is used, the pre-soak temperature can be in the range of 10° C. to 150° C.

After exposing the water insoluble pre-treated ligno-cellulosic biomass to vacuum conditions, enzymatic hydrolysis is conducted on the pre-treated material. The result is an increased yield of glucose and often xylose after the enzymatic hydrolysis when compared to a composition which has not been exposed to vacuum conditions.
