    Abstract: A combinable nucleic acid pre-processing apparatus includes a sample transfer chamber transferring a sample from a sampling tube to a nucleic acid extraction kit, a nucleic acid extraction chamber performing a nucleic acid extraction of the sample in the nucleic acid extraction kit for obtaining a nucleic acid extract, an assay setup chamber preparing reagents and transferring reagents and the nucleic acid extract to an assay plate, and at least two bridging modules respectively disposed between the sample transfer chamber and the nucleic acid extraction chamber and between the nucleic acid extraction chamber and the assay setup chamber. The sample transfer chamber, the nucleic acid extraction chamber and the assay setup chamber are separated and operated independently. Three chambers are connected through the bridging modules, so the nucleic acid extraction kit can be sequentially moved in the sample transfer chamber, the nucleic acid extraction chamber and the assay setup chamber.

The closure is engaged with the tube for enclosing the opening. The partitioning member is disposed in the tube and includes a blocking portion and a position-limiting portion. The blocking portion is disposed close to the opening and covers a portion of the opening so as to leave another uncovered portion as an entrance for passing the sampling swab therethrough. The position-limiting portion is connected with the blocking portion and extended into the tube, so as to limit the sampling swab in a space corresponding to the entrance inside the tube after the sampling swab passes through the entrance.

    Abstract: A nucleic acid extraction method includes the following steps. Firstly, a mixture of a sample and a lysis buffer is provided. The pH of the lysis buffer is lower than 7. Then, the mixture of the sample and the lysis buffer is transferred to a nucleic acid catching chamber containing chitosan coating material, so that nucleic acid of the sample is bound to the chitosan coating material. Then, the chitosan coating material is washed with at least one wash buffer, thereby removing residual protein and cell debris. Then, the chitosan coating material is eluted with an elution buffer while heating the chitosan coating material, so that the nucleic acid is separated from the chitosan coating material. The pH of the elution buffer is higher than 7.
