    Abstract: The present disclosure provides a P450 cytochrome enzyme for andrographolide synthesis and its application, belonging to the field of bioengineering. The present disclosure uses Saccharomyces cerevisiae CEN.PK2-1D as a host, and implements knockout of ROX1 and GAL80 genes on the genome, and integrative expression of GGPP synthase encoding gene and CPS diterpene synthase encoding gene at ROX1 site; and implements free expression of ApCPR and CYP71A8 and CYP71D10 both with truncated signal peptides, successfully constructing recombinant S. cerevisiae, and achieving de novo synthesis of 3,15,19-Trihydroxy-8(17),13-ent-labdadiene-16-oic acid. Compared with the blank, a response value of a product peak reaches 1.9*106, and this strategy provides necessary reference for analyzing biosynthetic pathway of andrographolide and using metabolic engineering to synthesize andrographolide and related derivatives thereof.

    Abstract: Provided is a method for the non-invasive detection of fetal genetic variations, comprising: firstly, estimating the percentage of fetal genetic material in a plasma sample of a pregnant woman by means of carrying out targeted sequencing on polymorphic sites on a reference genome and then performing allelic copy counting for each polymorphic site; and then, carrying out allelic copy counting on the polymorphic sites on a target genome or a target to be detected, and using a goodness-of-fit test or a relative distribution diagram of allele counts to detect whether the target to be detected in the sample has any variation at the chromosomal level, sub-chromosomal level or single genetic site level. The method is suitable for simultaneously detecting chromosomal euploidy variations, micro-deletion and micro-duplication variations at the sub-chromosomal level and variations at the short sequence level in the plasma sample of the pregnant woman, and has good development and application prospects.

The recombinant S. cerevisiae can be used for synthesizing carminic acid by taking self-synthesized acetyl-CoA and malonyl-CoA as a precursor. On this basis, OKS, cyclase, aromatase, C-glucosyltransferase and monooxygenase relevant to carminic acid are integrated to a high copy site, which can remarkably improve the yield of carminic acid. The yield of carminic acid can be increased to 2664.6 ?g/L by optimizing fermentation conditions, and the fermentation time is shortened significantly.

    Abstract: A method for predicting an air quality index (AQI) based on a fusion model proposes a differential fusion seasonal prediction model (DF-SPM) based on a random forest (RF) model and a convolutional neural network (CNN)-long short-term memory (LSTM)-attention (CLA) model. This method uses the optimal threshold interval (OTI) search algorithm to search and learn the OTI of four seasons during the prediction process, and obtains the final prediction results according to the predicted values of RF model and CLA model. The fusion model combines the prediction advantages of two independent models, and fully considers the seasonal and periodic characteristics of AQI, so as to accurately search OTI in different time periods with the seasonal scale, so as to achieve higher prediction accuracy. The OTI strategy of fusion model is superior to the single threshold strategy, which can extract the historical fluctuation characteristics of AQI and achieve higher prediction accuracy.

The devices and methods may include training a predictor based on test results using a supervised machine learning algorithm.     Abstract: The disclosure is about a method capable of realizing the preparation and in-situ separation of the oligomeric ricinoleate, which uses the ricinoleic acid as raw material, and uses a protonic acid-type ionic liquid as a catalyst to cause the dehydration and esterification reactions between ricinoleic acid molecules. By continuously distilling out the generated water under a reduced pressure, the oligomeric ricinoleate with a polymerization degree of 2 to 10 is obtained.
