    Abstract: A sugar cane plant cell comprising a recombinant nucleic acid molecule is provided, the recombinant nucleic acid molecule comprising a nucleotide sequence of interest, a promoter and the nucleotide sequence of SEQ ID NO:1, wherein the promoter is downstream of and in operative association with the nucleotide sequence of SEQ ID NO:1 and upstream of and in operative association with the nucleotide sequence of interest, and the nucleotide sequence of interest is expressed at a level at least about 6 times greater than the level of expression of said nucleotide sequence of interest in a control. Additionally, a method of increasing the expression of a nucleotide sequence of interest in a sugar cane plant cell using the recombinant nucleic acid molecule of the invention is provided.

    Abstract: Compositions and methods for increasing the expression and/or accumulation of cellobiohydrolase enzyme in the vacuoles of plant cells are provided. The method involves targeting the enzyme to the vacuoles through the use of a barley polyamine oxidase (BPAO) vacuole sorting signal peptide. Plants transformed with an expression construct encoding the vacuole sorting signal peptide operably linked to the cellobiohydrolase enzyme direct expression of the polypeptide to the vacuoles of the plant cells. Transgenic plants, seeds, and plant tissues, and plant parts are provided. Downstream uses of transgenic plants or plant material expressing the constructs of the invention include agronomical and industrial uses, for example, human food, animal feed, biofuel, industrial alcohol, fermentation feedstocks, and the like.

    Abstract: Compositions and methods for targeting polypeptides to plastids are provided. Compositions comprise plastid transit peptides as well as nucleotide sequences encoding such transit peptides and variants thereof. Compositions further comprise DNA constructs comprising a nucleotide sequence encoding the plastid transit peptide operably linked to a nucleotide sequence encoding a polypeptide of interest. These DNA constructs find use in expression and targeting of the polypeptide of interest to a plastid. Compositions also comprise expression cassettes, vectors, transformed plants, transformed plant cells, and stably transformed plant seeds wherein a polypeptide of interest is targeted to a plastid by the plastid transit peptide of the invention.
