    Abstract: A method and system of operating a microscopy system are disclosed. A first image first image of a sample acquired using a first imaging configuration is received. A second image is developed from the first image, wherein the second image is associated with a second imaging configuration. A sequence of image processing steps is applied to the first image to develop a first classification of first objects represented in the first image and to the second image to develop a second classification of second objects represented in the second image. A score associated with the second imaging configuration is developed that represents a difference between the first classification and the second classification. The image acquisition time or component requirements of operating the microscopy system is less when operated using the second imaging configuration than when operated using the first imaging configuration.

In an exemplary method, immersion fluid may be delivered onto a front end of an objective of the imaging system to form one or more boluses. Each bolus may have a rounded outer surface intersected by an optical axis defined by the objective. At least a portion of each bolus may be illuminated. Light may be detected from each illuminated bolus to create detection data. A value representing a size of each bolus may be determined using the detection data. Delivery of the immersion fluid may be adjusted, if needed, based on the value(s) determined.

The planar light guide is illuminated with one or more LED strips at side(s) of the light guide. A transparent or semi-transparent front cover is sealed over the LED strips and planar light guide with a rectangular seal that may have a circular cross section that partially lies within a channel formed in the bottom support. The light guide is backed with a reflector and one or more diffusers may be inserted between a top side of the light guide and the front cover, which is held in place with an inset frame that fastens to the bottom support.
