Publication number: 20240285727Abstract: Described herein is a novel, mitochondrial encoded, open reading frame that leads to the production of a new mitochondrial peptide called SHMOOSE. SHMOOSE is a 58-amino-acid peptide and, within its open reading frame, contains a genome-wide significant small nucleotide polymorphism (SNP) that markedly increased risk for Alzheimer's disease, brain structure, brain gene expression, and cognition. SHMOOSE increased neuronal-type cell survival and protected against amyloid beta toxicity. Metabolomic studies revealed a role for the peptide in energy optimization, whose dysfunction and dysregulation leads to cell death in physiologically notable regions of the brain in neurodegenerative diseases such as Alzheimer's and Parkinson's.
