    Abstract: An analysis approach used to evaluate base-call ambiguity and quality for a selected sequence. The methods described herein may be adapted to automated procedures for sequence acquisition, evaluation, and sample identification. In particular, the disclosed methods may be adapted to sequence identification techniques such as forensic analysis to provide an automated approach to data interpretation thereby reducing or eliminating the need for detailed investigator review. Implementation of these automated methods desirably provides a means by which to improve the speed and accuracy of the analysis as compared to conventional methods and may be used to improve identification throughput especially in large or complex sequencing projects.
