    Abstract: The present disclosure provides target nanodrugs comprising nanocarriers, such as nanodiscs and/or liposomes, encapsulating a therapeutic agent. The nanodrugs may be conjugated to a targeting antibody, such as for delivery of the nanodrug across the blood brain barrier. The nanodrugs may comprise anti-retroviral therapy. Further provided herein are methods for the treatment of a disease or disorder by administering the target nanodrugs, such as for the treatment of HIV.

In one embodiment, the nanoformulated drug is a retroviral drug. In one embodiment, the nanoformulated composition comprises a nanocarrier with one or more incorporated drugs. In an exemplary embodiment, the drug is efavirenz (EFV). In a further embodiment, the nanocarrier is associated with an agent for targeting microfold cells (M-cells). In a specific embodiment, the targeting agent binds to an M-cell marker known as glycoprotein 2 (GP2). Via this mechanism, the nanodrug is targeted toward GALT.

    Abstract: The subject invention provides materials and methods for treating diseases affecting the central nervous system (CNS) and/or other viral reservoir organs utilizing nanoscopic diamond particles, i.e., nanodiamonds (ND), loaded with therapeutic agents of interests. In one aspect, the subject invention provides a composition for treating a subject's brain and/or other organs acting as viral reservoirs, the composition comprising a plurality of ND particles measuring less than 10 nm in size, wherein the ND particles are loaded with at least one therapeutic agent of interest. In another aspect, the subject invention provides methods of treating disorders affecting the CNS such as, for example, the brain and other viral reservoir organs such as, for example, lymph nodes and gut-associated lymphoid tissues (GALT), utilizing the drug delivery system comprising a plurality of ND particles as provided herein.
