    Abstract: Novel compositions and methods are described for stimulating a patient's immune system to tumor cells which bear a gp72 tumor-associated antigen and thereby eliciting a tumoricidal effect. Anti-idiotypic human monoclonal antibodies and idiotopic regions thereof to primary antibodies which bind selectively to an antigenic determinant of gp72 are provided to mediate the humoral and cellular components of the patient's immune system. The immunogenic agents find use both in vivo and in vitro.

    Abstract: The present invention provides novel compositions and methods useful in the modulation or selective suppression of host immune responses to an immunogen of interest, particularly to immunogens which are exogenous antigens or allergens. Subject compositions include antibody, antibody derived, and antibody-like molecules of primary antigen reactivity with respect to the immunogen of interest. Antibodies or antibody-like or antibody-derived molecules include antibody fragments such as Fab, and complementarity determining region peptides (CDRs) which may be grafted into a framework region of any species, particularly human. They also include human antibodies, derived from sensitized human lymphocytes produced by cell fusion with heterohybridomas, or by DNA cloning and expression. Other compositions include T cell receptor (TCR) molecules, obtained either from T cell clones or hybridomas or as purified TCR preparations.

    Abstract: Novel methods and compositions are provided for the enhancement of the biodistribution of immunoconjugates useful in the diagnosis and treatment of a variety of conditions including cancer in many of its forms. The compositions of the present invention provide for enhanced bioavailability of immunoconjugates for the most part by blocking mammalian cell surface receptors present on cells of the reticuloendothelial system, especially in tissues responsible for the elimination of waste products and blood filtration. Such tissues include the liver, spleen, and kidneys. The compositions are administered in conjunction with an immunoconjugate in a pharmaceutically acceptable vehicle and may be provided in kits for convenient administration.

According to the present invention, MoAbs defining epitopes on either a tumor associated glycoprotein antigen of about 72 kD m.w. or on carcinoembryonic antigen conjugated to a ribosomal inhibiting protein, or the like, are employed either alone or in combination as cytotoxic agents in the treatment of various cancers including, but not limited to, colorectal carcinoma, ovarian carcinoma and osteogenic sarcoma. For some of these compositions, an enhanced or potentiated efficacy is observed when the conjugates are administered along with a related, unconjugated monoclonal antibody.Hybridomas XMMCO-791 and XMMCO-228 were deposited with the A.T.C.C. on Aug. 14, 1986 and given A.T.C.C. Accession Nos. HB 9173 and HB 9174, respectively. Hybridoma XMMBR-B14 was deposited with the A.T.C.C. on Jan. 14, 1987 and given A.T.C.C. Accession No. HB 9308.
