    Abstract: A condition-dependent, multiple target delivery system providing multifunctional, stimuli-sensitive pharmaceutical carriers is disclosed. The delivery system simultaneously carries on its surface various active moieties. The system is multifunctional and possesses the ability to switch on and switch off certain functions when necessary, for example, under the action of local stimuli characteristic of the target pathological zone (e.g., increased temperature or lowered pH values, which are characteristic of inflamed, ischemic and neoplastic tissues).

    Abstract: The invention provides composition and methods for delivering a composition according to the invention comprising a wild-type (wt) mitochondrial DNA (mtDNA) molecule to a mammalian cell. The wt-mtDNA molecule is a functional mtDNA molecule that includes the entire mitochondrial genome. The wt-mtDNA molecule in the composition of the invention has a mitochondrial leader sequence (MLS) peptide attached to facilitate the uptake of the mtDNA molecule into a mitochondrion of the mammalian cell. The delivery of the wt-mtDNA/MLS peptide complex is used as a general replenishment therapy for conditions attributed to mtDNA defects, independent of the specific defect in the mtDNA.

    Abstract: The invention is directed to an improved drug delivery system that includes a micelle, comprising polyethylene glycol and a lipid component, and a pharmaceutical agent dispersed in the lipid component. The delivery system may also include a targeting ligand. The micelle delivery system of the invention is capable of stabilizing, inter alia, poorly soluble pharmaceutical agents and increasing their delivery efficacy. Appropriate pharmaceutical agents useful in the system of the invention include anti-inflammatory agents, agents for photodynamic therapy, anti-tumor agents, anti-neoplastic agents, anti-metastatic agents, and imaging agents, as well as hydrophobized derivatives thereof. Specifically, the pharmaceutical agent can be porphyrin, chlorine-6-trimethyl ester, tamoxifen, paclitaxel, 1,3-bis(2-chloroethyl)-1-nitrosourea, camptothecin, ellipticine, rhodamine, dequalinium, diphenylhexatriene, vitamin K3, diethylene triamine pentaacetic acid, or a functional derivative thereof.

    Abstract: The preparation and use of a transducing polypeptide (TP)— lipid vesicle complex having a small proportion of positively charged (cationic) lipids in the make-up of the lipid vesicle, e.g., liposome, for safe and efficient intracellular delivery of therapeutic agents, such as proteins, DNA, small molecules and/or other drugs, into a cell of a higher organism, in vitro or in vivo is disclosed. The delivery system of the invention results in increased efficacy of intracellular delivery of such agents, bypassing the endocytotic pathway of intracellular delivery while at the same time minimizing the toxicity of the delivery system towards the recipient cells.
