Publication number: 20220113268Abstract: A low-cost photothermal biosensing method and apparatus for the quantitative genetic detection of pathogens such as MTB DNA on a paper hybrid device using a thermometer. First, DNA capture probes were simply immobilized on paper through a one-step surface modification process. After DNA sandwich hybridization, oligonucleotide-functionalized gold nanoparticles (AuNPs) were introduced on paper and then catalyzed the oxidation reaction of 3,3?,5,5?-tetramethylbenzidine (TMB). The produced oxidized TMB, acting as a strong photothermal agent, was used for the photothermal biosensing of MTB DNA under 808 nm laser irradiation. Under optimal conditions, the on-chip quantitative detection of the target DNA was readily achieved using an inexpensive thermometer as a signal recorder. Illustrative embodiments do not require any expensive analytical instrumentation, but can achieve higher sensitivity and there are no color interference issues, compared to conventional colorimetric methods.Type: ApplicationFiled: October 5, 2021Publication date: April 14, 2022Inventors: XiuJun Li, Wan Zhou

Publication number: 20210172940Abstract: A type of microfluidic platform, photothermal bar-chart chip (PT-Chip), uses on-chip nanomaterial-mediated photothermal effect as a tunable microfluidic driving force to drive ink bar-charts in a visual quantitative readout fashion. The photothermal bar-chart pumping performance can be adjusted remotely by tuning the irradiation parameters, without the need to change any on-chip parameters. The PT-Chip enables a POC visual quantitative diagnostics, by forming nanomaterial-mediated photothermal effects-driven bar-chart microchip for visual quantitative immuno-sensing. In this immunoassay, biomolecules are visually quantified by directly reading the distance that fluids move on the PT-Chip.Type: ApplicationFiled: December 4, 2020Publication date: June 10, 2021Inventors: XiuJun Li, Guanglei Fu, Wan Zhou
