    Abstract: A hand-held spinning centrifugal microfluidics system is provided for quantifying protein levels in biological samples. The system includes a spinning device and a centrifugal microfluidic chip disk having a hydrophobic upper layer and a hydrophilic bottom layer. The upper layer incorporates a reagent-loading chamber connected to multiple radially arranged reaction chambers through radial side-tubes. Each reaction chamber includes a venting and sample-loading opening for receiving biological samples. The radial side-tubes are constructed with a deliberate geometric transition to enhance local hydrophilicity and generate capillary pressure that prevents premature reagent flow before spinning. During operation, a colorimetric dye flows from the reagent-loading chamber into the reaction chambers. An illumination device provides uniform lighting, and an image acquisition module captures images of the resulting colorimetric reactions.
