    Abstract: The present disclosure relates to use of a Clostridium ghonii spore combined with pembrolizumab in cancer treatment. It is found for the first time that the Clostridium ghonii spore combined with pembrolizumab can significantly improve a curative effect of colon cancer and reduce a dose of the pembrolizumab, and thus is efficient and low-toxic. Oncolysis by Clostridium ghonii can affect immunogenicity of a tumor microenvironment (TME) by various ways, converts an immunosuppressive state of the TME into an immune-activated state, adjusts the immunosuppressive TME, and breaks an immune tolerance. An optimal combination of the Clostridium ghonii spore and the pembrolizumab thoroughly removes about 20% of mouse tumor tissues. A benefit range of patients with tumors treated by a PD-1 antibody is expanded. The combination even has an obvious curative effect on patients failed the treatment by the PD-1 antibody.

    Abstract: Methods and systems for budgeted and simplified training of deep neural networks (DNNs) are disclosed. In one example, a trainer is to train a DNN using a plurality of training sub-images derived from a down-sampled training image. A tester is to test the trained DNN using a plurality of testing sub-images derived from a down-sampled testing image. In another example, in a recurrent deep Q-network (RDQN) having a local attention mechanism located between a convolutional neural network (CNN) and a long-short time memory (LSTM), a plurality of feature maps are generated by the CNN from an input image. Hard-attention is applied by the local attention mechanism to the generated plurality of feature maps by selecting a subset of the generated feature maps. Soft attention is applied by the local attention mechanism to the selected subset of generated feature maps by providing weights to the selected subset of generated feature maps in obtaining weighted feature maps.

    Abstract: In the field of biomedical technology, a PIWI-interacting RNA piR-hsa-211106 is used to prepare a targeted therapeutic drug for inhibiting proliferation of lung adenocarcinoma cells. A mechanism is as follows: after the PIWI-interacting RNA piR-hsa-211106 is constructed into an agonist or a transformant, the agonist or the transformant inhibits a tricarboxylic acid cycle process by down-regulating an expression of a pyruvate carboxylase, inhibits an energy metabolism, promotes apoptosis of the lung adenocarcinoma cells, thus inhibits growth of the lung adenocarcinoma. The PIWI-interacting RNA piR-hsa-211106 directly acts on a target site and does not produce toxic and side effects and an off-target phenomenon. Large amounts of analysis and in-vivo and in-vitro experiments show that the PIWI-interacting RNA piR-hsa-211106 has high credibility and a remarkable treatment effect, and provides a new research direction for anti-tumor therapy of lung adenocarcinoma.

First, chitosan is dissolved in acetic acid solution; a chitosan-based thermosensitive hydrogel is printed by a 4D bioprinter and lyophilized after solvent extraction, to obtain lyophilized chitosan; subsequently, aqueous ?-sodium glycerophosphate solution is prepared with ultrapure water and ?-sodium glycerophosphate, and then aqueous carboxymethyl chitosan solution is prepared with ultrapure water and aqueous ?-sodium glycerophosphate solution are charged into and mixed well with aqueous carboxymethyl chitosan solution to prepare a mixture; finally, the lyophilized chitosan is crosslinked with the mixture to obtain the 4D chitosan-based thermosensitive hydrogel. With scientific and reliable principles thereof, the present invention solves a problem that conventional thermosensitive hydrogels have uneven pore sizes, and improves the entrapment efficiency and ability of limbal stem cells.
