    Abstract: The present disclosure discloses a method for rapidly determining survival condition of spores. The method includes the following steps: S100: performing sterilization treatment on the spore; S200: preparing a spore suspension with the spore subjected to the sterilization treatment; S300: dripping an appropriate amount of the above prepared spore suspension onto a solid medium, wherein 4-MUG is contained in the solid medium; S400: observing the solid medium under an excitation light source after the spore suspension fully infiltrates into the solid medium; and S500: determining the number of viable spores according to the number of the observed fluorescent spots on the solid medium. The method is based on an ?-glucosidase test method and can rapidly determine activity of the spore and judge activity of a single spore.

    Abstract: The present disclosure discloses a method for measuring two-dimensional distribution of an industrial irradiation dose based on sprouting lag time of spores, and provides a film for measuring distribution of an industrial irradiation dose. The method includes the following steps: (1) manufacturing the film: spreading a layer of spore suspension flat on a plane of a carrier, and evaporating a suspension solvent to fix a layer of spore pixel surface on the plane of the carrier; covering the carrier with a glass lid to form a sprouting cavity to cover the spore pixel surface within the sprouting cavity; and providing the glass lid with a filling hole configured to add a sprouting agent; (2) obtaining a calibration curve; and (3) measuring the irradiation dose.

    Abstract: TA preparation method for off-the-shelf human umbilical cord-derived mesenchymal stem cells includes culturing an isolated umbilical cord tissue blocks to obtain P0 cells; carrying out passage on the P0 cells twice to obtain P2 cells, and cryopreserving the P2 cells; the cryopreserved P2 cells are subjected to thawing and passage twice to obtain P4 cells, and the P4 cells are cryopreserved; the cryopreserved P4 cells are subjected to thawing and passage once to obtain P5 cells, and the P5 cells are sequentially digested, washed and resuspended to obtain the human umbilical cord-derived mesenchymal stem cells. The present invention also adopts cryopreservation method to develop a cryopreservation preparation of human umbilical cord-derived mesenchymal stem cells. The cryopreservation preparation is convenient for storage and can be directly administered intravenously after thawing without changing the liquid, avoiding possible contamination risk during dispensing, and is more conducive to clinical use.
