    Abstract: The present disclosure relates to a device and a method for reinforcing a round section wood beam by the combination of a prestressed Fiber Reinforce Plastic (FRP) sheet and a high strength steel wire rope. The device includes an FRP sheet adhered to a bottom surface of a log beam in the length direction. A middle supporting piece is mounted in the middle of the log beam. An end part reinforcing anchoring piece is mounted at each of two ends of the log beam. FRP hoops are adhered to the log beam and are located between the middle supporting piece and each of the end part reinforcing anchoring pieces at intervals in the length direction. High strength steel wire ropes with both ends connected to the corresponding end part reinforcing anchoring pieces are respectively arranged on the two sides of the middle supporting piece.

    Abstract: Conventional gene transformation requires tissue culture, and some elite lines have very low transformation efficiency in tissue culture. The disclosure relates to methods of in planta transformation. In some aspects, an axillary meristem of a plant is wounded and contacted with a transformation agent. The wounded axillary meristem is then regenerated and treated with a selection step, resulting in transformed tissue that can produce transgenic seeds.

    Abstract: The present invention is directed to a genomic DNA extraction reagent and method for improved extraction of DNA from biological tissue. The extraction reagent of the invention is mixed with disrupted biological tissue to form a DNA extraction solute which is incubated in a DNA extraction step. The extraction reagent includes an alkali component to maintain the DNA extraction solute at a pH of about 10 to 14 substantially throughout the extraction step. The extraction solute is centrifuged to clarify the supernatant. The supernatant containing the extracted DNA is diluted with a neutralizing buffer resulting in a high throughout method of generating high quantities of high quality DNA. Major PCR inhibitors are managed with the unique chemical combinations of the DNA extraction reagent designed and optimized for extraction of DNA from plant tissue and cells.
