    Abstract: The present disclosure relates to fluorescent biosensors and methods of use thereof. In particular, provided herein is a genetically encoded fluorescent indicator (GEFI) comprising a circular-permuted fluorescent protein (cpFP) and an inhibitory molecule bound to the cpFP. In the basal state, the inhibitory molecule prevents fluorescence from the circular-permuted fluorescent protein. Upon conformational change and/or cleavage of the bond between the inhibitory molecule and the cpFP, the cpFP is disinhibited, thereby permitting fluorescence from the cpFP. The biosensors described herein may be used in methods for detecting a variety of cell-signaling events.

    Abstract: Provided is a method for synthesizing a chiral amine compound. A transaminase is used to transaminate a ketone substrate under the action of an amino donor, to obtain the chiral amine compound; and the conserved amino acid sequence region of the transaminase at least includes a region 1 (MAGLWCVN) and a region 2 (YNTFFKT). With the transaminase with the specific conserved amino acid sequence region to synthesize a large sterically hindered chiral amine, the enzyme catalytic reaction volume is small, the synthesizing route is short, the product yield is high, a high-cost noble metal is not required for catalysis under the synthesizing conditions, three wastes are reduced, and the production cost is saved.

    Abstract: A computing system automatically manages error reports. Each error report specifies an error that occurred within a subsystem of the computing system. A received error report is added into a root cause grouping. Each root cause grouping contains error reports having error types traceable to a same root cause. A deployment time at which the subsystem corresponding to the error report was deployed within the computing system is determined. A severity score for the root cause grouping is generated as a function of the deployment time. The severity score inversely correlates to a time period length between the deployment time and the occurrence time of the error. The root cause grouping is assigned to a ranked error container of a plurality of ranked error containers based on the generated severity score. Each ranked error container contains root cause groupings having severity scores within a specified score range.

    Abstract: A control method of mixed traffic flow on freeway ramp based on controllable connected and autonomous vehicles (CAVs) is provided. A ramp is divided into a normal driving section, a vehicle platoon formation section and an accelerating and merging section. A vehicle platoon is formed by a leading CAV and human-driven vehicles (HDVs). Time interval [tmin, tmax] for the vehicle platoon to completely reach the merging point S is calculated. CAVs on the main lane and ramp are cooperatively controlled, and a merging gap is reserved for the ramp vehicle platoon. The vehicle platoon is allowed to accelerate and merge into the main lane. By means of the Internet-of-Vehicle (IoV) technology, the traffic situation on the main lane and downstream merging zone can be obtained in advance, and speeds of the CAVs are cooperatively controlled to lead the ramp vehicles to safely merge into the main lane.
