    Abstract: The present invention relates to a method of producing a polypeptide having ?-hexosaminidase activity, comprising the steps of a) providing a yeast cell comprising a polynucleotide encoding a polypeptide having ?-hexosaminidase activity and having an amino acid sequence being at least 95% identical to the amino acid sequence shown in SEQ ID NO: 1, b) cultivating said yeast cell under conditions which allow for the production of the polypeptide, and c) obtaining the polypeptide produced in step b). The method includes providing forged and NDT tested rotor discs and machining the discs for weld seam preparation. The weld seam preparation has an inner narrow TIG welding gap and an adjoining outer SAW welding gap. The method further includes stapling the discs on top of each other; and checking the run-out of the stapled discs relative to each other and as a whole and, if necessary, adjusting the staple. In addition the method includes melting the root of the weld without weld filler using TIG welding; increasing the weld height by narrow gap TIG welding with parent metal weld filler to allow tilting of the rotor in horizontal position; tilting the rotor in horizontal position; finalizing the welding by filling the outer SAW welding gap using SAW welding; and checking the welds of the rotor by NDT using ultrasonic testing.

    Abstract: A volume-adjusting insert for sample vessels comprises an elongated body with a septum seal on one end and a seal surface on the outside of the body. A through-chamber in the body provides a receiving chamber for a hypodermic needle inserted into the septum. The seal surface of the insert defines a reduced-volume sample chamber comprising the through-chamber and the vessel chamber portion of the sample vessel below the insert, or alternatively, below the seal surface. Sample fluid injected into, or withdrawn from, the insert communicates with the through-chamber and the lower vessel chamber. The device is particularly useful in converting multi-well microplates to low sample-volume vessels for automated sampling and testing, and allows the hydraulic pressure generated by a hypodermic needle to transport the sample fluid through the sample vessel and out of a bottom-extraction element.
