    Abstract: Herein, we provide genetically engineered immune effector cells, among other cells, which express CAR and secret a bispecific “trap” protein co-targeting a checkpoint protein and TGF-? or TGF-? receptor, so as to improve the antitumor immunity of the immune effector cells. Compared with conventional CAR-T cells and CAR-T cells secreting a polypeptide checkpoint inhibitor, the provided genetically engineered immune effector cells CAR-T cells with “trap” protein secretion attenuate inhibitory T cell signaling, enhance T cell persistence and expansion, and improve effector functionalities and resistance to exhaustion. In a xenograft mouse model, CAR-T cells with “trap” protein secretion significantly enhanced antitumor immunity and efficacy. Methods of using these genetically engineered cells, as well as using polynucleotides encoding the CAR and the “trap” protein, are also provided, for example, as a therapy against solid tumors.

    Abstract: A method for preparing organopolysiloxane resins, including the steps below: (1) using an alkoxysilane, a capping agent and an acidic compound as raw materials; (2) mixing all the raw materials uniformly, and the capping agent, the alkoxysilane and the acidic compound have a mole ratio from 0 to 40:1:0.2 to 5; (3) performing a polycondensation reaction at a temperature of 40-150° C. in 2-20 h; (4) adjusting the reaction product to neutral, and obtaining a product of organopolysiloxane resin after purification. Preparation of organopolysiloxane resins with the present application method has benefits on less environmental pollution, low energy consumption, efficient productivity, high yield of silicone resin with excellent performance.

    Abstract: Compositions are provided including NK cells that express chimeric antigen receptors (CARs) specific to CD19 and Her2 and a plurality of cell surface-bound multilamellar liposomal vesicles loaded with one or more anti-cancer therapeutics at an effective amount for inhibiting or killing tumor cells without causing toxicity to the NK cells. Methods of using these compositions to treat a subject with tumor are also provided, including administering an effective amount of the CAR-engineered NK cells, where an effective amount of anti-tumor therapeutics are delivered in particles (e.g., crosslinked multilamellar liposomal vesicles) that are bound to the surface of these CAR-engineered NK cells, without causing toxicity to the carrier NK cells.
