Abstract: Provided herein are trispecific binding proteins comprising four polypeptide chains that form three antigen binding sites that specifically bind one or more target proteins, wherein a first pair of polypeptides forming the binding protein possess dual variable domains having a cross-over orientation, and at least two of the antigen binding sites specifically bind an interleukin-4 (IL-4) polypeptide and an interleukin-13 (IL-13) polypeptide, respectively. In some embodiments, the third antigen binding site specifically binds an OX40L polypeptide, an interleukin-17 (IL-17) polypeptide, or a natural killer group 2D (NKG2D) polypeptide. The disclosure also relates to nucleic acid molecules, expression vectors, host cells, compositions (e.g., pharmaceutical compositions), methods of production, methods of use, and uses related thereto.
