    Abstract: RNA editing tools for use in systems designed to measure RNA in vivo and manipulate specific cell types are disclosed herein. An RNA sensor system comprising a) a single-stranded RNA (ssRNA) sensor comprising a stop codon and a payload; optionally wherein the ssRNA sensor further comprises a normalizing gene; and b) an adenosine deaminase acting on RNA (ADAR) deaminase; wherein the sensor is capable of binding to a ssRNA target to form a double-stranded RNA (dsRNA) duplex that becomes a substrate for the ADAR deaminase; wherein the substrate comprises a mispairing within the stop codon; and wherein the mispairing is editable by the ADAR deaminase, which editing can effectively remove the stop codon so as to enable translation and expression of the payload. A method of quantifying ribonucleic acid (RNA) levels using the RNA sensor system is also disclosed.

The concrete is formed by mixing and stirring base stocks, aggregates, admixtures, external additives and water. The base stock of the concrete is 17.4-17.5 parts of Portland cement; the aggregates include 38.9 parts of basalt with aggregate size of 5-10 mm and 33.1-33.2 parts of basalt medium sand; the admixtures are 1.9-1.95 parts of silica fume or fly ash, and further including 0.23-0.24 part of polycarboxylate water reducer and 1.34-1.35 part of sulfate corrosion-resistant liquid preservative. Optimized proportion method: according to the corrosion characteristics of sulfate and corrosion environment parameters, determine the composition and proportion of basic samples and comparison samples, make and cure sample components, test the deep components of the samples, and obtain the optimal composition and proportion according to the test results.

    Abstract: To program intercellular communication for biomedicine, it is crucial to regulate the secretion and surface display of signaling proteins. If such regulations are at the protein level, there are additional advantages, including compact delivery and direct interactions with endogenous signalling pathways. A modular, generalizable design is provided called Retained Endoplasmic Cleavable Secretion (RELEASE), with engineered proteins retained in the endoplasmic reticulum and displayed/secreted in response to specific proteases. The design allows functional regulation of multiple synthetic and natural proteins by synthetic protease circuits to realize diverse signal processing capabilities, including logic operation and threshold tuning. By linking RELEASE to additional novel sensing and processing circuits, one would be able to achieve elevated protein secretion in response to “undruggable” oncogene KRAS mutants.
