  • Publication number: 20220349017
Abstract: The present disclosure provides, among other things, primers and probes for detecting shedding of an AAV construct or fragment thereof in a subject. In some embodiments the primers are selected to generate an amplicon that comprises (i) a first strand comprising (1) a nucleotide sequence corresponding to the forward primer, and (2) a nucleotide sequence corresponding to a portion of the AAV construct or fragment thereof, (ii) a second strand comprising (1) a nucleotide sequence of the reverse primer, and (2) a nucleotide sequence that is complementary to the portion of the AAV construct or fragment thereof, or (iii) a combination thereof, where the portion of the AAV construct or fragment thereof comprises a nucleotide sequence that spans a junction between a regulatory element and the therapeutic gene of interest.

  • Patent number: 10564075
Abstract: Traditionally, enriching or isolating rare circulating cells from a sample has proven difficult as rare circulating cells can be heterogeneous, which limits the use of techniques based on biological properties (e.g., cell surface markers), and can be flexible, which hinders techniques based on physical properties (e.g., diameter size). Methods disclosed herein are capable of enriching or isolating rare circulating cells are minimally impacted by heterogeneity or flexibility of rare circulating cells, and therefore, overcome disadvantages observed with prior techniques. For example, a method for enriching or isolating rare circulating cells from a sample of a subject including fixing one or more rare circulating cells in a sample to create a fixed sample, and isolating the one or more rare circulating cell from the fixed sample using a size exclusion technique is disclosed.

  • Publication number: 20160299129
Abstract: Methods for implementing a biosensor system for highly sensitive and highly specific detection of a biological analyte are disclosed. The detecting methods include interacting the analyte to at least two or more sensitive elements (SE) and two or more signal transducing/amplifying molecules (TA) that form a stable sensitive-element-signal transducer/amplifier complex (SETAC) for further detection. Sequential format and concurrent format of the methods are disclosed. Multiplex format and automated format of the methods are disclosed. Biosensor systems and assay kit implementing these methods are disclosed.
