    Abstract: The invention relates to cell biology and regenerative medicine, specifically to a culture method and application for porcine limbal epithelial stem cells. The method involves pretreatment of cells, coating of culture dishes, centrifugation, culturing, passaging, and identification by immunofluorescence staining. The cultured cells are applicable in repairing corneal diseases, including limbal stem cell deficiency. During repair, the cells can be combined with biological materials to form a stem cell engineering carrier for implantation at the injury site. The separation and culture method is simple, yields a large number of cells, and is suitable for use with porcine eyeballs. Limbal epithelial stem cells cultured in vitro form distinct cell clusters, referred to as stem cell islands. The obtained cells exhibit phenotypes including BMI1+, ?Np63?+, CK14+, and ABCG2+.

    Abstract: The invention relates to bioinformatics technology and specifically to an analysis method of a spatial transcriptome-based 4D-printed stem cell scaffold for enhancing diabetic skin injury healing. Spatial transcriptome sequencing is performed on tissues from both the PBS group and the combined treatment group. Cell clustering and annotation identify main cell types, followed by visualization of their spatial distributions in both groups. The proportions of various cell types are statistically quantified and plotted. Cell subsets exhibiting significant differential gene expression are enriched and verified. Using a 4D-printed stem cell scaffold combined with a piRNA inhibitor and stem cell therapy, the method reveals spatial distribution and dynamic cellular changes during wound healing. It identifies precise cellular locations and gene expression patterns within tissues, providing critical insights into the biological processes by which biomaterials promote diabetic injury repair.

PEMSCs are sourced from uterine tissue of pigs, providing a streamlined process yielding a substantial quantity of cells. Cultured PEMSCs exhibit CD34?, CD11b?, CD44+, and CD105+ phenotypic markers and can differentiate into osteocytes, adipocytes, and chondrocytes in vitro. The method employs type II collagenase to degrade extracellular matrix collagen and DNase I to break down DNA, reducing cell aggregation. The interaction between piR-hsa-164586 and MYH9 can up-regulate the expression of MYH9 in NSCLC cell lines, the verification experiment process was confirmed by Transwell experiment after knockdown of piR-hsa-164586 and MYH9 respectively.

The invention uses dolomite as raw material, and the amount of magnesium hydroxide produced by magnesium oxide at a specific hydration temperature and hydration time can be ignored.
