    Abstract: Provided is a mutant hydroxyphenylpyruvate dioxygenase (HPPD) polypeptide, an encoding gene thereof and a use thereof. The mutant HPPD polypeptide retains the activity of catalyzing the conversion of hydroxyphenylpyruvate acid into homogentisic acid or homogentisate, and the sensitivity to HPPD inhibitor herbicides is lower than that of original unmutated HPPD. On position 372 corresponding to the amino acid sequence represented by SEQ ID NO: 1, the mutant HPPD polypeptide comprises the following mutations: F372A, F372G, F372V, F372P, F372S, F372T, F372C, F372M, F372Q, F372D or F372 deletion. The described mutant can provide plants with high tolerance to HPPD inhibitor herbicides, and can be used to cultivate plants that are tolerant to HPPD inhibitor herbicides.

    Abstract: An application of MAL33 gene deletion in improving the tolerance of Saccharomyces cerevisiae to inhibitors in a lignocellulose hydrolyzate is provided. The tolerance of the present MAL33 gene-deleted Saccharomyces cerevisiae strain to acetic acid is greatly improved, and the tolerance of the Saccharomyces cerevisiae strain to other typical inhibitors and H2O2 in the lignocellulose hydrolyzate is also improved. The lag period of the Saccharomyces cerevisiae strain in a glucose and xylose medium (YPDX) with 3.5 g/L acetic acid is shortened by 24 h. The fermentation period of the Saccharomyces cerevisiae strain to produce ethanol through co-utilization of glucose and xylose is shortened by 20 h. The growth of the Saccharomyces cerevisiae strain in a glucose and xylose medium (YPDX) with a mixed inhibitor and the ethanol production of the Saccharomyces cerevisiae strain through the co-fermentation of glucose and xylose are superior to those of a control strain.

The strain and screening strategy used in the present invention provide technical reference and basis for the further breeding of a Saccharomyces cerevisiae strain which has a superior fermenting property and is produced by the second generation fuel, ethanol.
