    Abstract: A methylation detection method and kit for genomic DNA or cell-free DNA molecules in a biological sample are provided. The method includes: obtaining a converted DNA molecule through a chemical and/or enzymatic reaction; performing a methylation-specific multiplex PCR, through multiple pairs of methylation-specific primers, on one or more methylated areas of one or more genes in the converted DNA molecule serving as a substrate, in order to obtain a PCR amplification product; and detecting the methylation levels of different fragments of the amplification product. The detection method can detect DNA methylation in multiple genes in multiple samples at the same time, requires only a small initial amount of DNA, is suitable for detecting methylated fragments having a small copy number, and has such advantages as high sensitivity, high specificity, high accuracy, a low detection limit, and low detection cost.
