    Abstract: Disclosed is a stereophotogrammetric method based on binocular vision, including the following steps: image acquisition, image correction and stereo matching are performed; cost matching and cost aggregation are performed on images of different sizes after correction; image segmentation is performed on the corrected image to determine edge pixel points of the object to be measured; and a pixel distance at an edge of the object to be measured is calculated to measure the size of the object. The method of the present invention enhances the matching accuracy of contour pixels of the object to be measured and improves the measurement accuracy.

A training set of images taken at unreduced and reduced power levels used to excite fluorescence during sequencing by synthesis is used to train a neural network to enable the neural network to recover enhanced images, as if taken at the unreduced power level, from unenhanced images taken at the reduced power level.

    Abstract: Methods of determining an ideal gain for a detector in a light detection system of a flow cytometer are provided. Methods of interest include irradiating a flow stream with light from a light source, incrementally increasing the gain of the detector such that the detector collects light from the flow stream at each of a plurality of successively increasing gains, obtaining a baseline noise level from the detector at each gain in the plurality of successively increasing gains, calculating a limit of detection (LoD) for each gain in the plurality of successively increasing gains, and assessing the calculated LoDs to determine the ideal gain. Systems and computer readable storage media for practicing the invention are also provided.
