The method includes: inoculating isolated mononuclear cells in a pre-coated culture flask or a culture dish for culture treatment; and performing activation and induction culture in a culture medium containing monoclonal antibodies and cytokines, to finally obtain NK cells. The coating is performed in a buffer containing anti-CD16 monoclonal antibody and IL-21.
