    Abstract: A primer set and a kit is used for simultaneous identification of a classical strain and a variant strain of a porcine epidemic diarrhea virus (PEDV). The primer set includes primer set I and primer set II; where primer set I comprises the nucleotide sequences set forth in SEQ ID NO: 1 to SEQ ID NO: 4; and primer set II comprises the nucleotide sequences set forth in SEQ ID NO: 5 to SEQ ID NO: 8. This approach not only helps avoid false negatives or false positives in the detection of PEDV variant strain infections, but also enables the identification between classical strain and variant strain in PEDV. This is of significant importance for understanding the prevalence of PEDV and for the prevention and control of PEDV infections.

    Abstract: Provided is a method for constructing a gE protein-deleted pseudorabies virus (PRV) strain using an adenine base editor (ABE) and use thereof. The method includes: designing an sgRNA sequence using the ABE with a start codon of the gE gene in a PRV as a target site, ligating an enzyme-digested fragment to a double-stranded DNA fragment with sticky ends to obtain a ligation product; and transforming the ligation product into a competent cell to allow plate screening and culture, selecting a resulting positive bacterial strain to allow expanded culture, and extracting a plasmid from a resulting positive bacterial solution; and transferring the plasmid into a target cell to allow the transfection for 24 h, collecting a resulting virus liquid, and centrifuging the virus liquid to collect a resulting supernatant.

    Abstract: The present disclosure provides a kit for detecting an African swine fever virus (ASFV), and provides a corresponding single guide RNA (sgRNA) with a nucleic acid sequence shown in any one of SEQ ID NO: 1 to SEQ ID NO: 5. The kit is based on loop-mediated isothermal amplification (LAMP)-clustered regularly interspaced short palindromic repeat (CRISPR)/Cas12b and can detect the ASFV in one tube at a constant temperature. The kit only needs to set one reaction temperature, does not open its lid midway, and has high sensitivity and specificity while showing no specificity to other swine viruses. The kit exhibits high efficiency and convenience, and does not rely on a large-scale experimental equipment. Compared with the traditional fluorescence quantitative PCR methods, the kit has a greatly improved sensitivity in detecting 1 copy/?L. The kit realizes visual detection by combining with colloidal gold test strip detection.

    Abstract: A three-level grid multi-scale finite element method for simulating groundwater flow in heterogeneous aquifers is proposed in present disclosure. The three-level grid refers to dividing the study region into coarse grid elements, then dividing each coarse grid element into medium grid elements, and finally dividing each medium grid element into fine grid elements, thereby improving the coarse-scale basis function construction method of the multi-scale finite element method. The new method of constructing a coarse-scale basis function by using the multi-scale finite element method itself instead of the finite element method is provided, constructing medium-scale basis functions on local medium grid elements, and using the medium-scale basis functions to construct a coarse-scale basis function in each coarse element, which can significantly improve the construction efficiency of the coarse-scale basis function.

During operation, the circuit uses a transformer-based balun to convert a single-ended input signal into a differential signal, wherein the transformer-based balun includes a transformer with a primary coil, which receives the single-ended input signal, and a secondary coil, which produces the differential signal. The transformer also includes a central compensation capacitor coupled between a center tap of the secondary coil and ground, wherein the central compensation capacitor acts to suppress common-mode components in the differential signal. Next, the circuit uses a pseudo-differential amplifier to convert the differential signal into a single-ended output signal, which has double the frequency of the single-ended input signal.
