Patent number: 11607458Abstract: Disclosed herein is a chemical delivery system having: i) a cargo compound comprising a first reactive moiety covalently bonded to a first enrichment moiety and a tethered cargo moiety, wherein the first reactive moiety is bonded to the tethered cargo moiety via a cleavable linker; and ii) a trigger compound comprising a second reactive moiety covalently bonded to a second enrichment moiety and a cargo-releasing moiety. The first enrichment moiety and the second enrichment moiety cause an increase in concentration of the cargo compound and the concentration of the trigger compound at a target site, causing a bimolecular reaction between the first reactive moiety and the second reactive moiety to form a cyclization precursor compound. The cargo moiety is then released from the cyclization precursor compound in a unimolecular cyclization reaction. Methods for treating conditions such as cancer, inflammatory conditions, and infections with the chemical delivery systems are also described.Type: GrantFiled: October 25, 2018Date of Patent: March 21, 2023Assignee: GEORGIA STATE UNIVERSITY RESEARCH FOUNDATION, INC.Inventors: Binghe Wang, Yueqin Zheng, Xingyue Ji, Robert Emuobonuvie Aghoghovbia

Patent number: 10751344Abstract: Carbon monoxide-releasing organic molecules are described herein. The molecules can be synthesized prior to administration (e.g., ex vivo) or formed in vivo. In those embodiments where the molecules are formed in vivo, reactants are administered under physiological conditions and undergo a cycloaddition reaction to form a product which releases carbon monoxide. In applying such reactions for therapeutic applications in vivo, the cycloaddition and CO release typically occur only under near-physiological or physiological conditions. For example, in some embodiments, the cycloaddition reaction and/or release of carbon monoxide occur at a temperature of about 37° C. and pH of about 7.4. Pharmaceutical compositions and methods for release carbon monoxide are also described.Type: GrantFiled: May 27, 2019Date of Patent: August 25, 2020Assignee: GEORGIA STATE UNIVERSITY RESEARCH FOUNDATION, INC.Inventors: Binghe Wang, Danzhu Wang, Xingyue Ji, Alexander Draganov, Chaofeng Dai, Krishna Damera, Didier Merlin, Emilie Viennois, Yueqin Zheng

Patent number: 10689335Abstract: The present invention provides methods of forming hydrogen sulfide. The methods include contacting a precursor compound with an unmasking agent; wherein the precursor compound comprises a hydrogen sulfide releasing moiety and a masked nucleophile; and wherein the contacting is conducted under conditions sufficient for cyclization of the precursor compound via lactone or lactam formation; thereby releasing hydrogen sulfide from the precursor compound. Hydrogen sulfide precursor compounds according to Formula I are also described, as well as methods for treating diseases and conditions using hydrogen sulfide precursors.Type: GrantFiled: April 4, 2016Date of Patent: June 23, 2020Assignee: Georgia State University Research Foundation, Inc.Inventors: Binghe Wang, Yueqin Zheng, Kaili Ji, Bingchen Yu, Zhixiang Pan
