    Abstract: The present invention provides in vitro methods for producing an endothelial cell with blood-brain barrier (BBB)-like properties. The methods include culturing an endothelial cell in a medium comprising a Wnt/?-catenin signaling activator and expressing one or more transcription factors in the endothelial progenitor cells for at least 2 days. The one or more transcription factors are selected from DACH1, DACH2, FLI1, FOS, FOXC1, FOXF1, FOXF2, FOXQ1, HES1, JUN, KLF2, KLF4, LEF1, MECOM, NR4A1, NR4A2, PPARD, TBX3, TSC22D1, ZIC2, ZIC3 and combinations thereof.
