    Abstract: A method for arterial endothelial-enhanced functional T cell generation is provided. In the method, arterial endothelial cells enhance functional T cell generation by promoting the generation of hematopoietic progenitor cells with T-lineage bias. The first stage of T cell differentiation from human pluripotent stem cells (hPSCs) is optimized, and it is found that hPSC-derived autologous arterial endothelial cells increase the T cell potential of hematopoietic progenitor cells. Moreover, the T cells generated by arterial endothelial cell priming share similar function to that of human peripheral blood T cells. hPSC-derived CD19-CAR-T cells have been verified to have tumor-killing effects both in vivo and in vitro. The established hPSC-T differentiation system would provide a valuable resource for chimeric antigen receptor T cell (CAR-T) therapy.

    Abstract: An approach is disclosed that provides personalized two-way communication with a uniquely identified object and an AI agent. Information about the object in a context associated with a requestor is received to access to an object knowledge base. The information is analyzed to determine a unique reference for the object. A selected set of the received information and the object categorization may be sent to an object knowledge base populated with AI configuration parameters tied to uniquely identified objects. The object knowledge base is searched for the unique reference to determine a registration assessment. The registration assessment is one of registered and not registered. When the registration assessment is determined to be not registered, the object is added to the object knowledge base. After receiving an AI connection from the object knowledge base, the context associated with the requestor is sent to the AI connection.

    Abstract: The present disclosure relates generally to systems and methods for determining an order of nucleotides of an RNA molecule. The method includes receiving liquid chromatography-mass-spectrometry (LC-MS) data of an RNA sample, filtering the LC-MS data based on mass, the filtering including removing masses smaller than a predetermined size, analyzing the filtered LC-MS data, to determine a plurality of RNA sequences, and reading-out an RNA sequence after determining no remaining valid nucleotides in the remaining LC-MS data. Analyzing the filtered LC-MS data includes determining a mass difference between at least two adjacent ladder fragments, and determining whether the mass difference is equal to a canonical nucleotide, or a modified nucleotide. The LC-MS data including a mass, retention time (RT), and volume. The RNA sequence including a sequence of each identified canonical nucleotide and any identified modified nucleotides.

The calculated hash value can be used to select an entry that defines how the packet is to be handled. The performance of the lookup operation can be monitored and if the hash calculation is resulting in excessive collisions or extra processing steps are needed in connection with the lookup operation, then the inputs to the hash calculation can be modified to attempt to improve the performance of the lookup operation.     Abstract: A method of improving quality of crowdsourced affective data based on agreement relationship between a plurality of annotators include receiving, by a processor, a collection of stimuli previously given affective labels by the plurality of annotators, executing, by a processor, an algorithm operative to perform the steps including constructing an agreement multigraph as a probabilistic model including a pair-wise status of agreement between the affective labels given by different ones of the plurality of annotators, learning the probabilistic model computationally using the crowdsourced affective data, identifying a reliability of each of the plurality of annotators based on the learned model, and adjusting the crowdsourced affective data by calculating the affective labels of each stimuli based on the identified reliability of each of the plurality of annotators, thereby improving the quality of the crowdsourced affective data.
