    Abstract: A protease with a function of degrading carotenoids of scallop is provided, which belongs to the technical field of genetic engineering. In particular, the present invention relates to the sequence and application of a carotenoid oxidase gene of scallop. The present invention analyzes the gene sequence, and verifies that the gene has function of degrading carotenoids of scallop scallop by RNA interference technology, and can be used to increase the carotenoid content of the scallop muscle of scallop scallop. The gene verified that the gene expression products can degrade carotenoids accumulated in bacteria. In addition, the RNA interference method provides a powerful tool for the study of scallop functional genes; the prokaryotic expression and protein purification methods provided can efficiently prepare oxidative lyases of carotenoid in scallop, so as to provide necessary experimental tools for in-depth research of the mechanism of shell carotene metabolism.

    Abstract: The present invention discloses a construction method for serial sequencing libraries of RAD tags, including the following steps: conducting an enzyme digestion reaction with DNA using endonuclease; ligating; enzyme-digested fragments with adaptors that contain restriction enzyme sites of SapI, featured base sequences for serial ligation of RAD tags, and universal sequences for the binding of amplification primers; conducting PCR amplification using a combination of biotin primers and general primers; collecting target PCR product by gel; amplifying again; and equally mixing and purifying; conducting enzyme digestion on the PCR products using the SapI enzyme and heating tags in series; purifying the long serial tags through gels and then conducting PCR.

    Abstract: The present invention relates to a method in the field of shellfish breeding techniques, for breeding a scallop population that have improved carotenoid content in the orange-red adductor muscles or other muscle tissues. This method includes the steps of selecting the rare individuals that have orange-red adductor muscles from natural scallop populations, growing the selected scallops, inducing the reproduction, artificially fertilization, and finally breeding a scallop population with orange-red adductor muscles which can be expanded for the cultivation on a large scale. Comparing to other known breeding methods, the present method does not use transgenic technologies to include introduce any exogenous genes, thus does not have any bio-safety and ethics issues. All the mutant scallops with orange-red adductor muscles are selected from natural or cultivated populations. After breeding for four generations, the obtained scallops have abundant carotenoid ingredients in their orange-red adductor muscles.
