    Abstract: Provided are compositions and methods useful for duplicating/amplifying a target fragment on a target DNA sequence such as a genome sequence. The editing system employs a pair of pegRNA which, by virtue of their targeting sites flanking the target fragment, extend the target fragment with reverse transcriptase (RT) templates included in the pegRNA. As the two RT templates at least include portions that are complementary to each other, they can form a duplex region which can then serve as starting point for DNA polymerase to synthesize a new strand for each strand of the target fragment, thereby duplicating the target fragment. Continue this process introduce amplification of this targeted sequence. Alternatively, this process can be done by combination of pegRNA/sgRNA or sgRNA/sgRNA. In the case of sgRNA/sgRNA in a PAM-out position, the RT enzyme and templates are not required.

    Abstract: Systems, devices, and methods for fabricating staircase structures in three-dimensional (3D) semiconductor devices are provided. In one aspect, a method includes providing a first deck comprising a first stack of first sacrificial layers and first isolating layers extending along a first direction. At least a part of the first deck is etched to form a first staircase structure. A second deck adjacent to the first deck along a second direction is provided, the second deck comprising a second stack of second sacrificial layers and second isolating layers extending along the first direction, the second sacrificial layers and the second isolating layers alternating with each other along the second direction. At least a part of the second deck is etched to form a second staircase structure. Contact structures extending through at least one of the first staircase structure or the second staircase structure along the second direction are formed.

    Abstract: A culture composition for expanding a TIL, containing IL-2, IL-7, IL-15, and an immune checkpoint antibody or an antigen-binding fragment thereof. In the culture composition, cell proliferation and activity are ensured while greatly reducing the used amount of IL-2, and IL-2 treatment is removed after infusing the TILs obtained using the culture composition into a patient so as to reduce side effects; moreover, no feeder cells are used in an expansion process, exogenous pollution is reduced and the manufacturing process is simplified, the manufacturing costs reduced, and the manufacturing time shortened.
