Abstract: Proto-antigen-presenting surfaces and related kits, methods, and uses are provided. The proto-antigen-presenting surface can comprise a plurality of primary activating molecular ligands comprising a major histocompatibility complex (MHC) molecule configured to bind to a T cell receptor (TCR) of a T cell and a plurality of co-activating molecular ligands each including a TCR co-activating molecule or an adjunct TCR activating molecule, wherein an exchange factor is bound to the MHC molecules. Exchange factors include, e.g., dipeptides such as GL, GF, GR, etc. Proto-antigen-presenting surfaces can be used to rapidly prepare antigen-presenting surfaces comprising one or more peptide antigens of interest by contacting the proto-antigen-presenting surface with one or more peptide antigens so as to displace the exchange factor. As such, the disclosure facilitates rapid evaluation of the immunogenicity of peptide antigens for activating T lymphocytes.

Abstract: An assay device allows reducing manufacturing variation, maintaining the measurement accuracy of the assay device at a high level, and enhancing the liquid control performance. The assay device of the present invention includes a microfluidic channel 2 allowing liquid to flow, an absorbing porous medium 3 disposed at a distance from one end 2a of the microfluidic channel 2, a separating space 4 disposed between the microfluidic channel 2 and the absorbing porous medium 3, and a housing space 5 housing the absorbing porous medium 3. The assay device further includes a lower member 20 which is an integrally molded article constituting a part of the assay device. The lower member 20 defines a lower portion 2b of the microfluidic channel 2, a lower portion 4a of the separating space 4, and a lower portion 5a of the housing space 5. The lower portions 4a, 5a of the separating space 4 and the housing space 5 are inclined downward toward one side from the other side in a flow direction.

Abstract: The invention provides a passive fluidics circuit for directing different fluids to a common volume, such as a reaction chamber or flow cell, without intermixing or cross contamination. The direction and rate of flow through junctions, nodes and passages of the fluidics circuit are controlled by the states of upstream valves (e.g. opened or closed), differential fluid pressures at circuit inlets or upstream reservoirs, flow path resistances, and the like. Free diffusion or leakage of fluids from unselected inlets into the common outlet or other inlets at junctions or nodes is prevented by the flow of the selected inlet fluid, a portion of which sweeps by the inlets of unselected fluids and exits the fluidics circuit by waste ports, thereby creating a barrier against undesired intermixing with the outlet flow through leakage or diffusion.
