Abstract: Compositions and methods using antibodies which are immunoreactive with specific apolipoproteins to determine the concentrations of lipoproteins such as HDL and LDL, and/or apolipoproteins in human blood, serum or plasma sample, are described. Monoclonal antibodies (MAbs) are described that specifically bind to epitopes present in apolipoproteins and lipoproteins, enabling rapid and reliable determinations of levels of specific blood lipoprotein and/or apolipoprotein levels, including Apo B-100, Apo A-I, Apo A-II, Apo C-III, and Apo E, and thereby determination of relative ratios of HDL and LDL and LpaI and LpaII. In a preferred embodiment, the compositions are strips of a solid phase material coated with one or more of the antibodies and are referred to herein as "dipsticks". The dipsticks specifically bind a lipoprotein or apolipoprotein when dipped into a protein sample.

Abstract: The invention relates to a process for the in vitro diagnosis of allergic, in particular also pseudo-allergic, immunological and environment-related disorders using live biopsy tissue samples. Also disclosed is a mobile incubation device for keeping the tissue samples alive and functioning. The samples can be, for example, skin or mucous membrane particles. The biopsy sample (14) once removed from the patient is placed in a temperature-controlled oxygen-containing incubating medium (16) and an allergen is introduced, triggering an immediate reaction (a so-called type 1 allergy). The secretion of immunoglobulin E (IgE) or a mediator is then determined qualitatively and/or quantitatively. Suitable mediators are histamine, tryptase, ECP, MPO, DAO, TPS, interleukins, prostaglandins or cytokines. The proposed mobile incubation device is provided with a temperature-controllable incubator for sample holders inside a housing. The sample containers can be filled with a nutrient solution.

Abstract: The present invention involves the identification of sphingoglycolipid species that are indicative of multidrug resistance in certain types of cells, including cancer cells. The association of multidrug resistance with the expression of certain sphingoglycolipids provides a new method for identifying multidrug resistant cancers. In addition, it has been determined that reducing the levels of certain sphingoglycolipids results in enhanced chemosensitivity of drug resistant cancer cells. This offers the opportunity to develop new treatments for multidrug resistant cancers.

According to the present invention, environmental samples to be tested are added to a pre-prepared physiological composition of bile salts and lipids. They are thoroughly mixed and then the resulting mixture is separated e.g. by centrifugation. The supernatant is then analyzed for the presence of contaminants and these concentrations are compared to the level of contaminants in the untreated samples. Abstract: A nucleotide sequence characteristic of Neisseria gonorrhoeae is disclosed. The sequence can be the basis for hybridization type, nucleic acid-based, rapid, in vitro diagnostic assays. The unique nature of the sequence makes it possible to clearly discriminate N. gonorrhoeae from other Neisseria species thus eliminating or substantially reducing the number of false positive readings. A 350 base pair N. gonorrhoeae DNA restriction fragment was cloned after subtractive hybridization to Neisseria meningitidis DNA. In further cloning experiments the sequences adjacent to the original 350 base pair fragment were determined. A portion of this sequence was shown to detect 105 of 106 N. gonorrhoeae strains and no other Neisseria species. In addition to use as detection probes, all or portions of the nucleotide sequence can be used as a ligand for the sandwich capture of N. gonorrhoeae sequences and as primers for in vitro amplification of N. gonorrhoeae sequences.

The micellar recognition system is provided in a single thin layer in which the recognition component is distributed homogeneously. Additional layers which may be provided about this layer, have an influence on the properties of the entire layer structure. On contact between the layer structure and the substance to be measured a recognition step takes place in the layer which is followed by a transducing step. In this way a measurement variable is produced by means of which information is obtained on the substance. Abstract: There is provided a process for the preparation of a highly reactive hydrophobic silica capable of lipophilic binding of lipids present in aqueous media, in particular body fluids. The hydrocarbon substituted silica utilized in the first step of this process may be prepared from hydrophilic silica, suitably pyrogenic silica. This is then treated with a volatile hydrocarbon substituted silicon compound. The next step of the process comprises treating the substituted hydrophobic silica with an alcohol. In order to enable the thus produced material to selectively adsorb lipids in the presence of protein, to the substantial non absorption of the proteins (other than lipoproteins), a further step is carried out. This comprises treating with aqueous ammonia or amines wherein substituents on the amine nitrogen may be the same or different.

Abstract: The present invention provides for the screening of candidate substances to identify active compounds that inhibit multidrug resistance (MDR). The expression of glucosylceramides has been determined to be a marker of MDR. By measuring glucosylceramide expression in cells exhibiting MDR, and the reduction in glucosylceramide levels in the presence of a candidate substance, the present invention provides for the identification of MDR inhibitory compounds.

  • Patent number: 5869537
Abstract: This invention encompasses a substantially homogeneous lipid chemoattractant released from stressed mammalian tissue which is a neutral lipid which is acid labile and stable to base and is stable in boiling water. This lipid recruits macrophages but not neutrophils to stressed tissue. The invention also encompasses a method for detecting injured tissue by detecting the presence of the above described lipid chemoattractant in body fluids such as urine, serum and saliva. The invention also includes a method for reducing recruitment of macrophages to injured tissue by reducing the amount of the above lipid chemoattractant or by blocking the interaction of this lipid chemoattractant with its macrophage binding site. The addition of this lipid chemoattractant to injured skin tissue promotes healing.

Abstract: A device (10) for determining the percent fat of a meat product such as beef or pork. The device 10 is presented in two designs, the first design uses a mechanical device in the form of a slide chart (12) and the second design employs an electronic device which is disclosed in three design configurations. The slide chart includes a stationary member (14) having a plurality of percent moisture windows (30) and percent fat windows (32) and a movable member (62) having a multiplicity of percent moisture figures (72) and percent fat figures (74). When a selected percent moisture figure (72) is located on the percent moisture window (30) the equivalent percent fat figure (74) is displayed on the percent fat window (32). The first design of the electronic device consists of a hand-held electronic calculator (90) which calculates the percent fat of the meat product when the percent moisture is known.
